CpG ODN/Mangiferin Dual Delivery through Calcium Alginate Hydrogels Inhibits Immune-Mediated Osteoclastogenesis and Promotes Alveolar Bone Regeneration in Mice

The immune system plays an important role in the skeletal during bone repair and regeneration. controlled release of biological factors from could facilitate optimize remodeling process through regulation activities cells. This study aimed to determine effect delivery immunomodulatory biologicals on Immunostimulatory cytosine-phosphate-guanosine oligodeoxynucleotides (CpG ODN) glucosylxanthone Mangiferin (MAG)-embedded microbeads were incubated with P. gingivalis-challenged splenocytes, or co-cultured RAW264.7 CpG ODN/MAG-containing regeneration was then tested vivo a mouse alveolar defect model. results demonstrated that MAG significantly antagonized gingivalis proliferation reduced live/dead cell ratio. After addition ODN + microbeads, anti-inflammatory cytokines IL-10 IL-4 upregulated day 2 but not 4, whereas pro-inflammatory cytokine IL-1β responses showed no difference at both timepoints. RANKL production by splenocytes TRAP+ formation cells inhibited microbeads. Alveolar bony defects, filled increased new after 4 weeks. In summary, this evaluated hydrogel-based regimen for local regenerate defects. combined treatment may promote anti-microbial/anti-inflammatory effects inhibition RANKL-mediated osteoclastogenesis.
